Binding Mechanism

Ceramic hydroxyapatite stands apart from traditional chromatographic resins due to its dual-site structure—Ca2+ and PO43-—which combines multiple protein binding mechanisms, including anion exchange, cation exchange, and metal affinity. Specifically, the PO43- site forms ionic bonds with positively charged proteins, demonstrating cation exchange traits, whereas the Ca2+ site interacts with negatively charged proteins via metal affinity or anion exchange with available carboxyl clusters. These two distinct mechanisms have positioned hydroxyapatite as a vital chromatographic resin in biopharmaceutical downstream processing, attracting growing interests in the production of antibodies, vaccines, nucleic acids, enzymes, and recombinant proteins.

Specifications

To meet the diverse needs of biological separation and purification, Pinshan Biotech has developed two types of ceramic hydroxyapatite (Type Ⅰ and Type Ⅱ). Additionally, three particle sizes (20μm, 40μm, 60μm) are provided for selection, ensuring that the products can adapt to different application needs of customers.

◆Ceramic Hydroxyapatite Type Ⅰ Resin:

Fully sintered at 400°C, forming uniform spherical particles.
With a high protein loading capacity, it is particularly suitable for the efficient separation of acidic proteins.


◆ Ceramic Hydroxyapatite Type Ⅱ Resin:

Fully sintered at 700°C, it also forms uniform spherical particles.
Compared to Type Ⅰ, it has a lower protein loading capacity but excels in resolving early elution proteins and nucleic acids.
With lower affinity for albumin, it is more suitable for purifying various types of immunoglobulins.
